Am taking a Holland America cruise in May to the Greek Islands with 5 other couples. Can any one suggest things to do in santorini and Mykonos?

Welcome to Cruise Critic. :)
You can see some of what HAL has to offer for excursions by going here
http://www.hollandamerica.com/cruise-destinations/ShoreExcursions.action
and selecting the port. (You do not have to enter the Destination.) For Santorini, see Thira.
I also suggest you find the Roll Call for your ship ( click here (http://boards.cruisecritic.com/forumdisplay.php?f=86) ) and see what others are talking about doing. You may find some organizing private tours.

If you don't mind driving rent a car in Santorini. Than you can drive all around the Island. It was the best way to get around.

I agree, you don't need a tour from the ship on Santorini.
When you get on the Island, take the Finicular up to the town, or if you are brave enough, you can take the donkey or walk up the steps, (it is a long walk), from there, the town opens up. A car rental is easy, and drive to see Oia, and to all of the other sights on the island.
When we were there last, we rented a Smart Car, (2 seat roller skate), it worked out really well. The speeds on the island are slow, but the island is really not that big. So rent a car and go exploring.
BTW, if you get there early, have breakfast on the island. Find some little roof top restaurant and have the Yogut & honey, it is fantastic.

There will be multiple HAL sponsored excursions to choose from. I think it would be difficult to go wrong with virtually any of these. Just remember, that Santorini is probably one of the most beautiful and picturesque places you will ever visit. Whatever you do, be sure you allow time to just walk around in the town and enjoy the spectacular scenery.

There are wonderful Minoan ruins on Santorini-well worth a visit. While in Mykonos, visit the nude beach (not everyone was naked - I certainly wasn't) but there are fantastic bars with music on them.
