Customs after Key West?

Not exactly sure.

On one of my Jewel of the Seas cruise, we missed Grand Cayman and was replaced with Key West. We were told that we would be passing through immigration in Key West, so we did all the immigration stuff on board (passports and everything) in Key West.

When we returned to Miami, all we had to do was give them the customs form as we exited the terminal. No checkpoint.

Just booked a four night on the enchantment of the seas. Was curious if we have to go through customs again in Miami after going through customs in Key West.

Saab4444 is correct. You don't clear Customs in KW. You'll clear Immigrations in KW and Customs when you return to Miami. It's 2 separate processes. People tend to think the 2 are one-in-the-same because they are done by the same agency (CBP).
