A day ticket in the hop on hop off buses (3 interlocking routes) is probably a good start though the service buses are incredibly cheap if you want to get to a specific venue.

 

You will be berthed within walking distance of the city centre.

 

My first choice if you want to go a little further afield would be MDina - about 20 mins away by bus.

We did Mdina/Valetta tour, and thoroughly enjoyed it in January on the Star.

The national war museum is worth a half day. Even the kids loved it. On the flip side - we did a snorkel trip with sea adventures - they pick up at the cruise port and it was fantastic.
